How to Use This Estimator

  1. Enter the sequence length (number of amino acid residues)
  2. Count and enter the number of degradation-prone residues (Asp, Asn, Met, Gln, Cys)
  3. Select any terminal modifications (acetylation, amidation)
  4. Indicate if the peptide is cyclized or contains D-amino acids

Degradation-Prone Amino Acids

Stability Classification

High Stability

Estimated plasma t₁/₂ > 12 hours. Suitable for once-daily or less frequent dosing. Few degradation-prone residues, protected termini.

Moderate Stability

Estimated plasma t₁/₂ 1–12 hours. Requires 2–4× daily dosing or depot formulation. Some degradation-prone residues present.

Low Stability

Estimated plasma t₁/₂ 15 min–1 hour. Requires continuous infusion or frequent dosing. Multiple degradation-prone residues.

Very Low Stability

Estimated plasma t₁/₂ < 15 minutes. Requires depot formulation, PEGylation, or D-amino acid substitution for therapeutic use.
