Semaglutide vs Orforglipron

Semaglutide and orforglipron represent two distinct pharmacological strategies for GLP-1 receptor agonism. Orforglipron (LY3502970) is a fully synthetic, non-peptide, orally bioavailable small molecule that acts as an allosteric agonist at the GLP-1 receptor. Unlike semaglutide, it:

  • Is not a peptide — it is a small molecule with a defined chemical structure
  • Does not require peptide engineering for metabolic stability
  • Achieves oral bioavailability through inherent physicochemical properties
  • Acts at a distinct allosteric binding site on the GLP-1 receptor

This non-peptidic mechanism eliminates the need for injection, refrigeration, or the absorption enhancers required by oral semaglutide.
